Cursusdoelen
Na afronding van de cursus is de student in staat:
·         verschillen in categorieën eiwitten te benoemen (oplosbare en membraangebonden)
·         kenmerken van primaire, secundaire en tertiaire eiwitstructuur te beschrijven
·         uit te leggen welke factoren primaire, secundaire en tertiaire eiwitstructuur bepalen
·         uit te leggen waarin oplosbare en membraangebonden eiwitten in primaire, secundaire en tertiaire eiwitstructuur verschillen
·         te beschrijven hoe primaire, secundaire en tertiaire eiwitstructuur geconserveerd zijn in de evolutie (incl. theorieën)
·         thermodynamische berekeningen uit te voeren aan eiwit-ligand complexen
·         effecten te beschrijven van de binding van geneesmiddelmoleculen aan eiwitten
·         uit te leggen hoe affiniteit en specificiteit van eiwit-ligandinteracties  variëren
·         te beschrijven hoe eiwit-eiwit- en eiwit-DNA-interacties tot stand komen
·         ultragevoelige eiwitsystemen te beschrijven en hoe cooperativiteit en allosterie daarin een rol spelen

·         te beschrijven hoe chaperones voorkomen dat eiwitten aggregeren
·         uit te leggen dat er intrinsically disordered proteins zijn die nooit vouwen maar wel een functie hebben
·         vanuit thermodynamische principes de stabiliteit van de DNA-duplex toe te lichten
·         te beschrijven hoe DNA-polymerase nieuwe baseparen selecteert voor DNA-replicatie en hoe replicatiefouten worden gecorrigeerd

Inhoud
The fate of living organisms is determined by the action of protein machines. In this course we introduce the molecular processes of life from the perspective of the protein. This way we study fundamental molecular concepts of life, which include the principles determining protein structures, homology of protein structures and function, allostery and the mode of action of chaperones. We discover the social life of proteins, they interact with macromolecules such as other proteins or DNA and small molecules such as ATP or drugs, but protein activity can also be modulated by chemical modification. Finally, we learn how all this, protein structure, modification and interaction allow proteins to control biological processes, including DNA replication and translation.
